摘要
针对数字全息显微术可以重构细胞相位.利用球面光波作为参考光的反射式数字全息显微术重构细胞相位的实验中,理论和实验证明所记录的光场中叠加有球面光场的相位,须减除后才能得到细胞的实际相位.采用不同的方法减除球面光场所得结果不同.对不同方法及结果进行了比较,结果表明:先测量没有细胞时光场相位,再计算细胞相位的方法,优于用拟合出球面波相位计算细胞相位的方法.
Digital holographic microscopy can be used to reconstruct the phase of cells. A device of reflecting digital holographic microscopy using spherical reference optical wave was introduced,and both theoretic analysis and experimental results show that the recorded optical filed added with a phase of spherical wave. In order to get the correct phase of cells, phase of the spherical wave must be subtracted. The method of experimental data processing was discussed, and the fresh onion cells was tested and both the phase and 3D information of cells are obtained.
