缺血再灌注大鼠心肌细胞钙含量与组织型纤溶酶原激活物活性的相关性

摘要 目的:探讨大鼠(Sprague Dawley,SD)心肌缺血再灌注心肌细胞钙含量与组织型纤溶酶原激活物(tissue-type plasminogen activator,TPA)活性的变化,以及两者之间的相关性。方法:采用在体大鼠心肌缺血-再损伤模型,恢复血流再灌注。实验分正常对照组和缺血1h再灌注1h组、缺血1h再灌注2h组、缺血2h再灌注1h组、缺血2h再灌注2h组4个实验组,每组各取10例测定心肌细胞钙浓度与TPA活性的变化,并进行相关性分析。结果:缺血再灌注后,大鼠心肌细胞钙含量与TPA活性随缺血和再灌注时间的延长而升高(P<0.01),两者之间呈显著相关性(r=0.9231,P<0.05)。结论:缺血再灌注可引起心肌细胞钙含量升高,从而导致心肌组织TPA活性的增高,引起心肌组织结构和功能的损伤。 Objectives To investigate the changes in cell calcium content and TPA in rat myocardium during ischemia reperfusion. Methods: The rat model of myocardial ischemia reperfusion was established. The experimental groups were separated into 4 groups , which were myocardial ischemia lh-reperfused 1h, ischemia 1h-reperfused 2h,ischemia 2h- reperfused lh and ischemia 2h-reperfused 2h. 10 cases in every group were selected randomly to measure myocardial cell's calcium content and the activity of TPA. Results: In myocardial ischemia-reperfusion model, the calcium content and the activity of the TPA in the cells were increased with the continuing time of the ischemia and the reperfusion(r= 0. 9231 ,P〈0. 05). Conclusion: Myocardial ischemia reperfusion cause the increase of the calcium content in the cell. It is responsible for the high activity of TPA which contribute to the damages to the construction and the function of the tissue.
