高效液相法测定调味品中5’-鸟苷酸二钠和5’-肌苷酸二钠 被引量:10

摘要 采用Inertsil ODS-3色谱柱(250mm×4.6mm,5μm);以0.02moL/L磷酸二氢钾溶液(pH4.5)为流动相,流速1.0mL/min,柱温25℃,检测波长250nm,同时分离了样品中5'-鸟苷酸二钠和5'-肌苷酸二钠。结果显示:在选定色谱条件下5'-鸟苷酸二钠和5'-肌苷酸二钠线性关系良好,加样平均回收率为98.6%~100.3%;相对标准偏差0.77%~1.67%。该方法精密度好,结果准确可靠,适合于调味品中5'-鸟苷酸二钠和5'-肌苷酸二钠的定量分析。 Inosine-5' -monophosphate (5' -IMP) and guanosine -5'-monophosphate (5' -GMP) in samples were separated successfully using an Inertsil ODS-3 column (250 mm×4.6 mm, 5 μm) when 0.02 mol/L monopotassium phosphate (pH4.5) was employed as the mobile phase with a flow rote of 1.0 ml/min at 250 nm and 25℃. A good linear relationship between 5'-GMP and 5'-/MP was obtained under above conditions. The average recovery rote was 98.6%-100.3% and the relative standard deviation ranged from 0.77%-1.67%. This method is accurate and reliable for quantity analysis for 5' -IMP and 5' -GMP in condiments.
