反相高效液相色谱法测定小鼠血清及肝脏氧化苦参碱毫微粒的浓度

摘要 目的建立测定小鼠血清及脏器中氧化苦参碱毫微粒浓度的方法。方法采用反相高效液相色谱(RP-HPLC)法。色谱柱为ZORBAXSB-C18(250mm×4.6mm,5μm),柱温35℃,流动相为20%乙腈-磷酸-三乙胺(50mmol/L的磷酸,70mmol/L的三乙胺和3.7mmol/L十二烷基磺酸钠组成,pH=3.0),流速1ml/min,检测波长220nm,进样量30μl。结果方法的平均回收率为97.95%(RSD=0.45%),测定血药浓度线性范围为1.31-42.08μg/ml(r=0.9999),肝脏匀浆中药物浓度线性范围为1.31-39.26μg/ml(r=0.9971)。结论本方法简便,快速,稳定,灵敏度较高,可用于氧化苦参碱毫微粒的血药浓度测定及动物体内分布的研究。 Objective To develop a method to determine the OMT - PBCA - NP in serum and tissue homogenates ( in some organs) of mice. Methods RT - HPLC method for the HPLC determination was performed using ZORBAXSB - C18 column. The mobile phase was 20% acetonitrile - phosphoric acid - triethylamine (50 mmol/L phosphoric acid, 70 mmol/L triethylamine and 3.7 mmol/L sodium dodecylsulphate (SDS) were contained, pH = 3.0). Flow rate was at lml/min, the detection wavelength was at 215 nm, the column temperature was 35℃ and The sample size was 30ul. Results The recovery of the method was 97.95% ( RSD = 0.45% ). The linear range was 1.31 -42.08μg/ml (r =0.9999) in mice plasma and 1.31 - 39.26μg/ml ( r =0.9971 ) in mice liver homogenates. Condusion The method is simple, rapid, stable and highly sensitive, and suitable for the investigation of OMT - PBCA - NP in plasma and the study of its distribution in animal body.
