摘要
目的研究己酮可可碱对乳酸盐腹膜透析液致人腹膜间皮细胞(HPMC)损伤和TGF-β1、FN分泌的影响。方法采用胰蛋白酶-EDTA消化法,从人腹膜组织中分离间皮细胞,并体外培养HPMC。采用自动生化分析仪检测培养液中乳酸脱氢酶(LDH)水平以示细胞损伤程度;ELISA法检测细胞培养液中TGF-β1、FN的水平。结果与单纯对照组相比,2.5%L-PDS致培养液中LDH水平明显增高(P<0.01);同时培养液中TGF-β1、FN蛋白质水平增加。在2.5%L-PDS刺激下,100μg/ml己酮可可碱与下调培养液LDH,提高MTT渗入量,并减少TGF-β1、FN蛋白质水平。结论己酮可可碱可拮抗乳酸盐腹膜透析液致HPMC损伤并下调TGF-β1和FN的表达。
Objective To investigate the effect of Pentoxifylline (PTX) on human peritoneal mesothelial cells' (HPMC) function, TGF-β1 and FN secretion induced by lactate-buffered peritoneal dialysis solutes. (LPDS) Methods Mesothelial cells from human omentum were isolated and cultured in vitro. Proliferation of HPMC was detected the concentration of lacetate dehydrogenase (LDH) in supematants were detected by automatic biochemical analyzor. TGF-β1 ,FN protein were determined by a sandwich enzyme-linked immunosorbent assay (ELISA). Results LDH levels of supematants increased and proliferation of HPMC was inhibited in L-PDS groups( P 〈 0.01). 2.5% L-PDS increased the cell's expression of TGF-β1, FN. 100ug/ml Pentoxifylline (PTX) caused an increase in HPMC proliferation, a decrease in LDH concentration , and inhibited TGF-β1,FN expression. Conclusion FIX can reverse the injury of L-DPS to I-IPMC and down regulate the expression of TGF-β1,
