摘要
目的研究大鼠局灶性脑缺血后肝细胞生长因子(HGF)表达的动态变化。方法采用尼龙线栓法制作大鼠局灶性永久性大脑中动脉闭塞模型,用免疫组织化学方法观察了缺血后3h、6h、24h、7d、14d时,HGF表达的动态变化。结果大鼠缺血后3h,缺血侧脑组织开始出现HGF表达,7d时明显增多,14d时密集表达。各时间点HGF的表达主要集中在梗死灶周围。结论脑缺血后可上调HGF在大脑的表达,HGF参与了缺血性脑损伤的病理发展及修复过程。
Objective To investigate the changes of hepatocyte growth factor (HGF)expression in rat brains following ischemia. Methods We made focal permanent cerebral ischemia model by occluding the middle cerebral artery with nylon line in rats, to investigate the changes of HGF expression in brain tissue by immunohistochemistry. Results It was found that HGF started to express at 3h after onset of MCAO,then gradually increased, extened at 7d, and became further marked at 14d after MCAO. HGF expression was mainly detected in the periphery of the ischemia area at every timepoint. Conclusion HGF expression is up- regulated by ischemia. HGF may takes part in the pathological and recovery process of ischemic stroke.
