摘要
目的探讨IL-1β对人冠状动脉平滑肌细胞(HCASMC)分泌妊娠相关血浆蛋白A(PAPP-A)的影响。方法培养HCASMC,至第5~7代时,分别加入20μg/L和0μg/L的IL-1β(0μg/L组为对照组),分别孵育2、4、8、24、36 h后,收集细胞培养上清液;采用IL-1β(0、5、20、4μg/L)刺激HCASMC 6 h后分别收集细胞和细胞培养上清液。用ELISA法检测细胞培养上清液内PAPP-A的表达量。结果IL-1β组PAPP-A表达量在2 h时开始增加,8、24和36 h时其浓度显著高于对照组(P均<0.05);随着IL-1β剂量增加,PAPP-A的表达量不断升高,其中20μg/L和40μg/L组的浓度均显著高于0μg/L组的浓度(P均<0.05)。结论IL-1β可使HCASMC分泌PAPP-A增加,并呈时间和剂量依赖性。
[ Objective] To observe the effect of IL -1β on secretion of pregnancy -associated plasma protein - A (PAPP- A) in human coronary artery smooth muscle cell(HCASMC). [ Medthods] Incubated HCASMC with 20 μg/L IL -1β ( IL -1β group) and 0μg/L IL -1β( control group) ,collected cuhttre media at 2, 4, 8, 24 and 36 h. Stimulated HCASMC with IL-1β(0, 5, 20, 40μg/L respectively) , collected culture media after 6 h. Measured the PAPP- A in cultural fluid by ELLISA . [ Results ] PAPP - A expression began to increase at 2 h in IL -1β group, and PAPP - A concentration was significantly higher at 8,24,36 h than that in control group( P 〈 0. 05 ). PAPP -A expression increased with the increasing of IL-1β concentration, and PAPP- A concentration of IL-1β(20μg/L and 40 p.g/L)group was significandy higher than that of control group(P 〈 0. 05). [ Conclusions] IL -1β can induce secreation of PAPP - A in HCASMC and in a time - and concentration - dependent manner.
