摘要
目的探讨慢速冷冻-快速复苏法对人胎儿卵巢组织超微结构的影响。方法将人胎儿卵巢组织分为新鲜组和冻融组,对冻融前后的胎儿卵巢组织超微结构进行比较。结果与新鲜组相比,冻融组卵巢组织中细胞的超微结构大部分保存良好。冷冻损伤在卵母细胞主要表现为胞浆内的空泡,线粒体水肿,偶见核膜的皱缩;在颗粒细胞主要为轻度的水肿和少量空泡;间质组织未见明显损伤。结论慢速冷冻-快速复苏法可以较好地保存人胎儿卵巢组织超微结构。
Objective To investigate the effect of the slow-freezing and rapaid-thawing to the uhrastructure of the human fetal tissues. Methods The human fetal tissues were divided into two groups :the fresh group and the frozen-thawed group. The ultrustructures of the two groups were compared. Results When compared with the fresh group, the majority ultrastructures in the frozen-thawed group general were well preserved. But it was found that a few follicles presented damage signs of the structure after frozen-thowed. The cryoinjury of uhrastructure of follicles in human fetal ovarian tissue was characterized principally as the vacuoles in cytoplasm, the edema of mitochondria. The shrinkage of nuclear membrane was seen occasionally. The most injuries of granular cells were slightly edema and few of vacuoles. There was no obvious damage in the interstitial tissue. Conclusions The protocol of slow-freezing/rapid-thawing is suitable for keeping human fetal ovarian tissue.
