NO释放调控剂(S,S)ZX-5对eNOS mRNA转录与eNOS蛋白表达的影响 被引量:1

摘要 目的考察(S,S)ZX-5对eNOS mRNA转录与eNOS蛋白表达的影响,从分子水平探讨(S,S)ZX-5促进一氧化氮(NO)释放的作用机制。方法用不同浓度的新型化合物(S,S)ZX-5刺激ECV30424h,采用RT-PCR半定量方法研究(S,S)ZX-5对ECV304eNOS的mRNA转录水平的影响;用Westernblot方法研究(S,S)ZX-5对ECV304eNOS蛋白的表达量变化影响。结果30mg.L-1(S,S)ZX-5使ECV304的eNOS mRNA转录量增加119%、使eNOS蛋白表达量增加62%;30mg.L-1(R,R)ZX-5的上述作用不显著。结论(S,S)ZX-5促进ECV304NO释放的作用机制是通过增加内皮型的NOS(eNOS)mRNA转录,提高eNOS蛋白的表达水平,从而提高了eNOS酶的活性。该研究为将(S,S)ZX-5开发为通过增加NO释放从而选择性改善脉络膜血流的药物提供实验依据。 Aim To investigate the effects of(S,S)ZX-5 and(R,R)ZX-5 on eNOS mRNA and protein levels.Methods Cell culture was used for the experiments in vitro.The effects of eNOS mRNA were investigated by RT-PCR.The effects of eNOS protein levels were investigated by Western blot.Results(S,S)ZX-5 in concentration of 30 mg·L^-1 up-regulated eNOS expressions at protein and mRNA levels significantly in cultured ECV304(P〈0.01).The effects of(R,R)ZX-5 was of no significant difference.Conclusion The mechanism of NO increases in ECV304 by(S,S)ZX-5 were up-regulation of eNOS expressions at protein and mRNA levels.The result provide experimental evidence for a new drug for prevention and treatment of AMD(age-related macular degeneration).
