亚砷酸诱导人骨髓间充质干细胞凋亡的研究

摘要 目的研究亚砷酸(ATO)体外对人骨髓间充质干细胞(MSC)凋亡的影响。方法在正常人髂后上棘穿刺获得MSC,体外培养传代,取第3代细胞鉴定后进行实验。纯化的MSC与1、5、10μmol/L的ATO共孵育,分别在6、12、24 h用HE染色观察细胞形态学变化,透射电镜观察细胞超微结构,流式细胞术(FCM)分析细胞凋亡情况。结果ATO可致MSC形态明显改变;不同浓度ATO对MSC作用24 h可见不同阶段的凋亡细胞;FCM检测表明,ATO可诱导MSC凋亡,该作用随ATO浓度和作用时间增加而增强。结论ATO可在体外诱导MSC凋亡,并呈时间和浓度依赖性。 [ Objective] To study the effect of arsenic trioxide (ATO) on the apoptosis of human bone marrow mesenchymal stem cell (MSC) in vitro. [ Methods] MSC was obtained from pricking posterior superior iliac spine of healthy volunteers. It was cultured and went down to posterity in vitro and subcultured for three generations. After being identified, the cells were cultured with different concentrations of ATO ( 1,5 and 10 μmol/L). After 6, 12 and 24 hours, the morphological changes of MSC were observed by HE staining, the ultrastructure of the cells was observed with electron microscopy, and cell apoptosis was evaluated by flow cytometry. [ Results] ATO could induce morphological changes of MSC. While the MSC were treated with ATO for 24 hours, apoptotic cells of different stages could be observed, Flow cytometry analysis showed that ATO induced apoptosis of MSC, and the role increased with the ATO concentration increasing and time prolonging. [ Conclusion] ATO can induce MSC apoptosis in vitro in time-dependent and concentration-dependent manner.
