活血化瘀方抗被动吸烟对大鼠红细胞膜骨架蛋白损伤的研究 被引量:1

“Huoxuehuayu” Decoction Against Injury of Erythrocyte Membrane Skeletal Proteins Caused by Passive Smoking

摘要 用被动吸烟的大鼠模型,研究活血化瘀方抗红细胞膜骨架蛋白(EMSP)损伤的作用。红细胞膜经SDS-聚丙烯酰胺凝胶电泳,用光密度仪540nm扫描分析各蛋白成分的质量分数(%)。结果:模型组红细胞膜与对照组相比,高分子量聚合物(HMW)明显增高[分别为(11.27±0.53)%、(1.80±0.42)%,P<0.01],带3蛋白明显减少[分别为(12.83±0.93)%、(21.33±0.78)%,P<0.01);而用药组红细胞膜除HMW外,带3蛋白、带4.2蛋白与对照组大致相同(均为P>0.05);模型组红细胞膜巯基总量较对照组及用药组明显减少(分别为P<0.01、<0.05),用药组与对照组差异无显著意义(P>0.05)。提示被动吸烟后红细胞膜蛋白质有大量交联聚合,而本活血化瘀方有抗被动吸烟所致EMSP损伤的作用。 The anti-injury effect of 'Huoxuehuayu' decoction on erythrocyte membrane skeletal proteins was studied with a rat model of passive smoking. After the sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of erythrocyte membrane was performed, the percent amount of every skeletal protein was quantitated by densitometry of the Coomassiie Brilliant Blue R250 stained gel at 540 nm. The results showed that the amount of high molecular weight (HMW) complex increased and the band 3, band 4. 2 protein decreased significantly in the model group erythrocyte membrane compared with the control group and the treated group (P<0.01). Except HMW the treated group was similar to the control group in the amount of them. The total sulfhydryl amount of erythrocyte membrane was reduced significantly in the model group (P<0.05), and there was no significant difference between the treated group and the control group (P>0.05). The results suggest that polymerization occurs with a lot of erythrocyte membrane proteins and'Huoxuehuayu' decoction can protect erythrocyte membrane skeletal proteins from injury caused by passive smoking.
