摘要 目的:观察抗凋亡基因GW112(Olfactomedin 4)在胃癌组织的表达水平及临床意义。方法:采用SYBR Green I实时定量RT-PCR检测25例胃癌组织及胃癌外周正常胃组织中GW112的表达,并分析其表达情况与临床病理资料的相关性。结果:GW112基因在胃癌组织的表达水平(M=0.12)显著高于正常胃组织(M=0.02,P<0.05),在有淋巴结转移的胃癌组织中的表达(M=0.23)显著高于无淋巴结转移的胃癌组织(M=0.04,P<0.05),在肿瘤TNM分期Ⅲ/Ⅳ期的胃癌组织(M=0.23)的表达显著高于Ⅰ/Ⅱ期(M=0.04,P<0.05);在年龄高(>60岁)患者的胃癌组织中的表达(M=0.25)高于年龄低(≤60岁)的患者(M=0.06,P<0.05)。在性别和分化程度上差异没有统计学意义(P>0.05)。结论:GW112基因在胃癌组织中高表达,与胃癌的侵袭和转移相关,可作为胃癌侵袭、转移的标志物。 Objective:To investigate the expression levels of antiapoptotic-related gene GW112 in gastric cancer and control tissues and its clinical significance. Methods: GW112 expression levels were detected by SYBR Green Ⅰ quantitative real-time PCR in cancer and normal tissues surrounding the cancer in 25 patients. Analysis was performed to study the correlation between the expression of GW112 and clinicopathological parameters. Results: Expression levels of GW112 were significantly higher in gastric cancer tissues than in normal tissues ( M =0.12 vs M =0.02, P 〈 0.05). The expressions of GW112 in the lymph node invasive group were significantly higher than that of non- lymph node invasive group( M = 0.23 vs M = 0.04 ,P 〈 0.05) . Overexpression of GW112 was also found more frequently in stage m/iv cancer group( M = 0, 23 )than in stage I/II group (M = 0.04) . In addition, the expressions of GW112 mRNA in gastric cancer tissues from elder patients ( 〉 60 year) were increased significantly compared with that in younger patients (≤〈60 year) (M = 0.25 vs M = 0.06). GW112 expression was not correlated with the sex and histological grades of cancer ( P 〉 0.05 ). Conclusion: GW112 was highly expressed in gastric cancer tissues. The expression levels were positively correlated with the degree of malignancy, suggesting its use as a biomarker for invasiveness and metastasis of grastric cancer.
