活血降糖饮对实验性糖尿病大鼠作用及机制 被引量:4

Effect and mechanism of Huoxue jiangtang yin on experimental diabetic rats

摘要 目的:观察活血降糖饮对糖尿病大鼠血糖、血脂、血浆胰岛素、胰高糖素、胰岛素敏感性指数及氧自由基等指标的干预作用。方法:实验于2001-03/08在广州中医药大学第一临床医学院糖尿病研究所实验室进行。①造模:取80只SD大鼠随机分为2组,一组65只腹腔注射链脲佐菌素诱发糖尿病,另一组15只腹腔注射柠檬酸缓冲液为正常对照组。②给药:将造模成功的56只大鼠随机分为模型空白组、玉泉丸组、优降糖组及活血降糖饮组4组,每组14只。模型空白组每日灌服3mL生理盐水,玉泉丸组每日灌服玉泉丸混悬液3.75g/kg,优降糖组每日灌服优降糖混悬液1.25mg/kg,活血降糖饮组每日灌服活血降糖饮水煎液(黄芪30g、生地20g,丹参30g,太子参30g,五味子15g,麦冬15g,淮山12g,黄精15g,丹皮12g,大黄16g,川红花12g,桃仁10g组成)0.01mL/g,治疗5周。③评估指标:5周后采血检测空腹血糖、血脂、过氧化脂质、超氧化物歧化酶、血浆胰岛素、胰高糖素,并计算胰岛素敏感性指数。结果:60只大鼠进入结果分析。①空腹血糖:造模后各组均较造模前升高(P<0.001),治疗后活血降糖饮组低于玉泉丸组,高于优降糖组[(13.22±6.88),(15.35±7.23),(10.28±6.75)mmol/L,P<0.05],3组均高于正常对照组。②血脂:活血降糖饮组和玉泉丸组三酰甘油及胆固醇低于优降糖组,高密度脂蛋白胆固醇高于优降糖组(P<0.05)。③血浆胰岛素:治疗后优降糖组高于造模后[(9.15±2.66),(4.61±1.55)mU/L,P<0.05],玉泉丸组与活血降糖饮组与造模后无差异。④胰高糖素:治疗后各组均明显下降(P<0.01),其中活血降糖饮组下降程度强于玉泉丸组,弱于优降糖组(P<0.05)。⑤胰岛素敏感性指数:治疗后玉泉丸组与活血降糖饮组均较造模后显著改善(P<0.01),优降糖组无明显变化。⑥治疗后,治疗各组血浆过氧化脂质水平降低,超氧化物歧化酶活性升高,与空白组比较差异显著(P<0.01),但仍未达到正常对照组水平(P<0.05)。结论:活血降糖饮对实验性糖尿病大鼠有降低空腹血糖、改善血脂、降低胰高糖素及增强胰岛素敏感性的作用。 AIM: To observe the interventional effects of Huoxue jiangtang yin on blood glucose, blood lipids, plasma insulin, glucagon, insulin sensitivity index and oxygen free radicals of experimental diabetes rats. METHODS: The experiment was undertaken at laboratory of Institute of Diabetes, First Clinical Medical College, Guangzhou University of Chinese Medicine from March to August 2001. (1) Totally 80 SD rats were divided into 2 groups randomly: 65 rats in group A were treated with streptozotocin (STZ)-induced diabetes by intraperitoneally; 15 in group B were treated with citrate buffer solution by intraperitoneally as normal control group. (2) The 56 rats were divided into 4 groups randomly: model blank group, Yuquanwan group, glybenclamide group and Huoxue jiangtang yin group with 14 in each group. Rats in the model blank group were given 3 mL saline by gastric perfusion every day. Rats of the Yuquanwan group were drenched suspension of Yuquanwan (3.75 g/kg) each day. Rats of the glybenclamide group were drenched suspension of glybenclamide (1.25 mg/kg) each day. Rats of the Huoxue jiangtang yin group were drenched liquid of Huoxue jiangtang yin (composed of 30 g astragalus mongholicus, 20 g rehmannia dride rhizome, 30 g salvia miltiorrhiza, 30 g heterophylly falsestarwort root, 15 g fructus schizandrae, 15 g ophiopogonis tuber, 12 g dioscorea batatas, 15 g solomonseal rhizome, 12 g cortex moutan, 16 g rhubarb, 12 g sichuan safflower and 10 g peach seed; 0.01 mL/g) for 5 weeks. (3) After 5-week treatment, fasting blood glucose, blood lipids, lipid peroxidation (LPO), superoxide dismutase (SOD), plasma insulin and glucagon were measured and the insulin sensitivity index was calculated. RESULTS: Totally 60 rats were involved in the result analysis. (1) The level of fasting blood glucose was increased after establishing models (P 〈 0.001 ). It was lower in the Huoxue jiangtang yin group than the Yuquanwan group, but higher than the glybenclamide group [(13.22±6.88), (15.35±7.23), (10.28±6.75) mmol/L,P 〈 0.05]. The levels in the three groups were all higher than that in the normal control group. (2) The levels of triglyceride and cholestedn in the Huoxue jiangtang yin group and Yuquanwan group were lower than that in the glybenclamide group. Level of high-density lipoprotein cholesterol was higher than that in the glybenclamide group (P 〈 0.05). (3)Plasma insulin was higher in the glybenclamide group after treatment as compared with that after establishing models [(9.15±2.66), (4.61±1.55) mU/L, P 〈 0.05]. There was no significant difference between Yuquanwan group and Huoxue jiangtang yin group after establishing models. (4)Glucagon was decreased obviously after treatment in each group (P 〈 0.01 ). The reduction of Huoxue jiangtang )tin group was higher than that in the Yuquanwan group, but lower than that in the glybenclamide group (P 〈 0.05). (5)Insulin sensitivity index was ameliorated markedly in the Yuquanwan group and the Huoxue jiangtang yin group after treatment as compared with that after establishing models (P 〈 0.01 ). There was no significant change in the glybenclamide group. (6)After treatment, level of plasma LPO was decreased but level of SOD was increased. There were significant differences as compared with the blank group (P 〈 0.01 ), but did not reach the level in the normal control group (P〈 0.05). CONCLUSION: Huoxue jiangtang yin may bnng down fasting blood glucose and glucagon, ameliorate blood lipids, and improve the insulin sensitivity in experimental diabetes rats.
