摘要
【研究目的】研究苏云金芽孢杆菌(Bacillus thuringiensis,Bt)伴孢晶体的形成及降解,为其生物学特性的系统研究和活性成分分析提供理论基础。【方法】将Bt菌株HD-1在牛肉膏蛋白胨固体培养基上30℃下培养24h,制成菌悬液后用日本电子JEM100-CX-II透射电子显微镜观察伴孢晶体的形成。采用五种溶解方法降解Bt菌株HD-1和LSZ9408伴孢晶体后,进行SDS-PAGE电泳。【结果】此研究的电镜观察表明BtHD-1在分裂过程中形成芽孢和晶体,晶体为大菱形。BtLSZ9408晶体含有130和65kDa的毒蛋白,HD-1晶体含有130、65和25kDa的毒蛋白。【结论】BtLSZ9408和HD-1伴孢晶体都包含CryI和CryII蛋白质毒素。
[OBJECT]To study the formation and decomposition of crystal of Bacillus thuringiensis (Bt), and supply theorctics basis for systmic study on its biological characteristic and analysis of its active compo- nent. [METHOD]Bt strain HD-1 was cultured on NA culture media for 24h under 30℃, and then was made into bacterial suspension and observed the formation of crystal with transmission electron microscope (JEM 100-CX-II). Five dissolution methods were used to decompose the crystals of Bt HD-1 and Bt LSZ9408, after that SDS-PAGE was conducted. [RESULTS] Bt Hd-1 formed spores and crystals during split process, and the shape of crystal was big diamond. Bt LSZ9408 contained 130 and 65kl)a toxic pro- teins, while Bt HD1 had 130, 65 and 25kDa toxic proteins. [CONCLUSION]Both of crystals of Bt LSZ9408 and HD-I contained CryI and Cryll type protein toxins.
