养血润肠方对实验性小鼠血虚型慢性功能性便秘结肠肌间神经丛5-HT和AchE的影响

The Affection of Yangxuerunchang Formula on 5-HT and AchE in Colonic Homogenate and Serum of Blood-deficiency Type Chronic Functionality Constipation Mice

目的:研究养血润肠方对实验性小鼠血虚型慢性功能性便秘结肠肌间神经丛5-HT、AchE的影响。方法:取健康小鼠72只,随机分正常对照组、模型组、乳果糖组和养血润肠方小、中、大剂量组。正常对照组和模型组给予生理盐水,其他各组分别给予乳果糖口服液和不同剂量养血润肠方。各组动物灌服药物14天后,处死,分离大肠,行免疫组化染色分析。结果:模型组5-HT、AchE神经递质的阳性表达的神经细胞数量较正常组明显减少,养血润肠方治疗各组均可使结肠肌间神经丛5-HT、AchE则有不同程度的增加,而乳果糖则没有增加结肠肌间神经丛5-HT、AchE阳性表达的作用。结论:养血润肠方有调节慢性功能性便秘小鼠结肠肌神经丛5-HT、AchE的作用。

Objective：To probe the affectiou of Yangxuerunchang formula（YXRCF） on 5-HT and Ache in Colonic Homogenate and Serum of Blood-deficiency Type Chronic Functionality Constipation （CFC）Mice. Method：Seventy-two healthy mice were divided into six groups randomly, i. e. normal group（NG）, model group（MG）, LOL group（LoG） and YXRCCF low（YIG）, middle（YmG）, high （YhG） dose group. NG and MG were given normal sodium,any other group were given LOL or different dose YXRCF. After intragastric infusing two weeks,all mice were killed, colon were taken, then immunohistochemical stain of frozen section were made, Result.YXRCF can increase the quantities of positive-cells of 5-HT and AchE in myemeric plexus of CFC modeled mice colon, LOL group＇ can＇t increase the quantiues of positive-cells of 5-HT and AchE in myenteric plexus of CFC modeled mice colon. Conclusion： YXRCF can adjust the content of 5-HT and Ache in colonic homogenate and serum of Blood-deficiency type chronic functionality constipation mice.
