摘要
【目的】DNA的提取是GMO(Genetically modified organism)作物检测中重要步骤,DNA的质量关系到GMO检测的成败。【方法】本研究以大豆油为材料,探索出一种快速、简便的提取大豆油DNA的方法。【结果】该方法能从10ml大豆油中提取0.4μg高纯度DNA,可用于PCR检测。提取市场上出售的十几种精练大豆油的DNA,针对35S、Nos、EPSPS和Lectin基因进行了PCR检测,分别扩增出不同的目的片段。【结论】大豆油中有残存的DNA碎片,在转基因大豆油中能扩增出外源基因的片段。本研究为大豆油的外源基因的检测提供了可行方法。
[ Objective] DNA extraction is an important step in GMC (Genetically modified crop) detection. [Method] In this paper we have developed a simple and rapid- method for extraction of DNA from soybean oil. [Result] A yield of 0.4 μg NA/10 rnl oil can be obtained. PCR amplification of sequences of 35S, Nos, EPSPs and an endogenous reference gene lectin in 11 market available brand of refined soybean oil was performed. Results showed that the sequence-specific fragments were amplified in all of the samples, demonstrating that the method could be applied for detection of transgene sequences in refined soybean oil. [Conclusion] There are some DNA fragments in soybean oil, and PCR amplification can detect the foreign DNA. This research provide a method to isolate and detect the foreign DNA in soybean oil.
