摘要
采用水解圈法筛选到一株产碱性木聚糖酶的嗜碱芽孢杆菌NT-16。该菌株产酶的最佳碳源和氮源分别是木糖和胰蛋白胨,其优化的产酶条件为:半纤维素2%,胰蛋白胨1%,Tween-800.1%,K2HPO40.1%,MgSO4·7H2O0.02%,pH值10.0,200r/m,72h,37℃。该菌株产生的木聚糖酶的最适pH为9.0。
An alkalophilic bacillus NT-16 was screened by assaying transparent zone on the selective medium. Xylose was the optimum carbon source and pancreas-poptone was the optimum nitrogen source. Results showed that the optimal condition for enzyme production was hemicelluloses 2%, pancreas-poptone 1%, Tween-80 0.1%, K2HPO4 0.1%, MgSO4·7H2O 0.02%, with 72h cultivation at pH 10.0 and 37℃. The enzyme revealed the high activity at pH 9.0.
