摘要 目的建立电纺纳米纤维为萃取介质,填装固相微萃取小柱,用于犬血浆中维拉帕米的提取净化,用高效液相色谱-紫外检测犬血浆中维拉帕米的浓度。方法采用Bea-gle犬为试验对象,口服单剂量的盐酸维拉帕米片(20mg·kg-1),0.1ml血浆样品经处理后注入固相微萃取小柱,50μl甲醇洗脱,收集洗脱液进液相色谱分析。结果维拉帕米的线性范围为50~10000μg·L-1(r=0.9999),最小检测限为25μg·L-1,提取回收率为88.1%(RSD=4.55%),精密度为4.15%~6.01%。结论此法简单方便,可替代液-液萃取法,能够用于犬血浆中维拉帕米的测定。 Aim A novel microextraction procedure based on micro-column packed with polystyrene electrospun nanofibers as the sorbents was presented coupled with high-performance liquid chromatography( HPLC-UV ) for analysis of verapamil in dog plasma. Method Verapamil tablet(20 mg·kg^-1 )was administrated orally to three Beagles. 0. 1 ml plasma sample was loaded onto the micro-column. The analyte was eluted with a small aliquot of methanol, which can be directly injected onto the assay system. Results Verapamil was validated in terms of linearity(γ =0. 9999 for concentrations ranging from 50 μg·L^-1 to 10 000 μg·L^-1 ), precision ( 4. 15%, 4. 85%, 6.01%, respectively at 100,500 μg·L^-1 and 2 000 μg·L^-1) ,sensitivity (25 μg·L^-1 for LOD) and average extraction recovery (88. 1% ). Conclusion The developed procedure was considered as a good alternative to the liquid-liquid extraction methods normally used for the determination of this compound in biological media.
