HPSE、MMP-9和E-cad联合表达对中老年非小细胞肺癌侵袭、转移及其预后的影响 被引量:1

The effects of HPSE,MMP-9 and E-cad on the invasion,lymph node metastasis and prognosis of middle and aged people with non-small cell lung cancer

摘要 目的研究中老年非小细胞肺癌(NSCLC)中HPSE、MMP-9、E-cad mRNA和蛋白的表达,探讨其与肿瘤侵袭性、淋巴结转移及预后的关系。方法应用实时荧光定量PCR和免疫组织化学方法检测NSCLC组织中HPSE、MMP-9、E-cad mRNA和蛋白表达。同时,对随访资料进行生存分析,绘制生存率曲线,探讨三者对NSCLC患者预后的影响。另外,通过细胞黏附能力实验测定硫酸多糖对NSCLC A549细胞黏附能力的影响。结果HPSE、MMP-9 mRNA和微血管密度(MVD)值在不同肺组织中的表达有显著性差异(均P<0.05)。HPSE、MMP-9、E-cad的表达在不同年龄、性别、组织学类型及分化程度间无显著性差异(均P>0.05),而在淋巴结转移与否组间有显著性差异(P<0.05)。患者术后生存期与HPSE、MMP-9及E-cad的表达强度显著相关(均P<0.05)。生存率曲线显示HPSE和MMP-9蛋白高表达者生存时间均短于低/无表达者,E-cad蛋白则与之相反(P=0.027,0.018,0.030,n=65)。HPSE和MMP-9的表达与MVD值均呈正相关(r=0.48;r=0.54)。细胞黏附能力实验显示实验组NSCLCA549细胞黏附率明显比对照组低。结论HPSE、MMP-9、E-cad表达可作为预测中老年NSCLC侵袭、淋巴结转移和预后的良好指标;三者共同促进了NSCLC微血管形成和肿瘤细胞转移;HPSE抑制剂硫酸多糖能够有效抑制NSCLCA549细胞的黏附能力。  Objective To investigate the expressions of HPSE,MMP-9 and E-cad in non-small cell lung cancer(NSCLC) and to analyze their correlation with tumor invasion,lymph node metastasis and prognosis.Methods Real-time fluorescence quantitative PCR and immunohistochemistry method were used to detect the expressions of HPSE,MMP-9 and E-cad in NSCLC to analyze the effects of three factors on the prognosis of patients with NSCLC by drawing Kaplan-Meier curve and the effect of Laminarin sulfate on the adhesion ability of lung cancer A549 cell.Results HPSE,MMP-9 mRNA and microvascular density(MVD) had a higher expressions in NSCLC tissue than those of adjacent tissue and normal lung tissue(P〈0.05).There were no significant differences among the expressions of HPSE,MMP-9 and E-cad and age,sex,histological type and differentiation(P〉0.05).There were statistical relationship between expressions of them and lymph node metastasis and prognosis (P〈0.05).The Kaplan-Meier curves showed that patients with low or no expressions of HPSE and MMP-9 had a longer survival time than those with high expressions of them,but E-cad was opposite(P=0.027,0.018,0.030,n=65).There were positive correlation between HPSE,MMP-9 and MVD(r=0.48;r=0.54).The cell adhesion ability test showed that the experiment group had a lower adhesion ability than control group.Conclusions The expressions of HPSE,MMP-9 and E-cad can be used as three useful markers of judging the biological behavior and prognosis of lung cancer.They have a congenerous effect on angiopoiesis of NSCLC and metastasis of tumor cell.Laminarin sulfate can decrease the adhesion ability of lung cancer A549 cell.
