不同时期大鼠整胚石蜡标本的制备

Preparing Method of Paraffin Wax Embedding Samples of Rat Embryo at Different Developing Stages

目的探讨不同时期大鼠整体胚胎石蜡标本的制备。方法收集胚龄11天至19天的大鼠胚胎,经4%多聚甲醛固定、酒精梯度脱水、浸蜡、包埋技术制备石蜡标本,光镜下观察胚胎组织结构。结果不同时段的整胚石蜡标本其组织器官的脱水、透明和浸蜡均较完全,镜下观察各组织结构完整,层次清楚。结论经本实验采用的制备流程,胚胎石蜡标本效果良好。

Objective To explore the preparing method of paraffin wax embedding samples of rat embryo at different developing stages.MethodsThe specimens of rat embryos at 11-19 embryonic day（E11-E19） were collected,fixed in 4% paraformaldehyde inPBS,subsequently dehydrated in series of alcohols and embedded in paraffin wax,then the tissular structure of the embryos was observed by using light microscope.ResultThrough the procedure mentioned above,The structure between different tissues were easily to be determined and no fragmentation was found in tissues under the light microscope.ConclusionThe method in this study is effective and stable,and the morphologic characters are preserved.
