实验性糖尿病大鼠动脉粥样硬化模型的建立 被引量:13

摘要 目的建立类似人类糖尿病并发动脉粥样硬化(AS)大鼠模型。方法选1月龄SD大鼠,先饲以高脂高糖饲料4周,促发胰岛素抵抗,继以小剂量STZ(30mg/kg·体重)腹腔注射,诱导建立糖尿病AS模型。结果注射STZ2周后大鼠血糖、甘油三酯、高密度脂蛋白、低密度脂蛋白及HDL-L/总胆固醇均显著高于空白组;形态学观察发现,模型组动脉出现典型粥样硬化病变。结论本实验方法建立的糖尿病伴动脉粥样病变大鼠模型,与人类2型糖尿病代谢特征相似,是研究人类糖尿病大血管病变较理想的动物模型。 ] Objective:To estable rat model of diabetes mellitus with arteriosclerosis that simulates the metabolic abnormalities of human disease. Methods: 1 month old Sprague Dawley rats were fed on a diet enriched in fat and glucose to induce insulin resistance , after 4 weeks, the rats were injected intrapertoneally with a low - dose streptozotocin (STZ) (30mg/kg·BW)to induce hyperglycemia with arteriosclerosis. Results: After the 2nd STZ injection, the rats demonstrated the blood glucose, TG, HDL - L, LDL - L and HDL - L/TC levels were significantly risen up . Moreover, the significant pathological changes of arteriosclerosis appeared in the model rats' arteries after 11 weeks. So the successful model was made. Conclusion: This model closely simulates the human diabetes mellitus with arteriosclerosis, this study provides the ideal animal model for investigating human diabctes with macroangiopathy.
