摘要
以‘大红’甜橙实生苗上胚轴为外植体,研究其离体培养和植株再生技术的结果表明,MS+1.0mg·L-16-BA的培养基诱导不定芽的效果最好,30d时诱导率达97.5%。外植体抗性筛选出最适kanamycin浓度为50mg·L-1。IBA和NAA都促进不定芽生根。1/2MS+4.0mg·L-1IBA和1/2MS+1.5mg·L-1NAA诱导根的效果最佳,生根率和根数分别达到76.92%、0.81和92.31%、1.88。
The technique of plant regeneration in vitro from epicotyl segments of ‘Dahong' sweet orange was studied. The results showed that the best medium for adventitious buds induction was MS+ 1.0 mg.L^-1 6-BA, the induction rate was 97.5% in 30 d. The suitable concentration of kanamycin from the selection was 50 mg.L^-1. Both IBA and NAA could accelerate the rooting of adventitious buds. The best media for the rooting were 1/2MS+4.0 mg.L^-1 IBA and 1/2MS+1.5 mg.L^-1 NAA. The rooting frequency and the average root numberper shoot were 76.92%, 0.81 and 92.31%, 1.88 respectively.
