摘要 目的研究中华补血草根提取物(the Limonium sinense Ktze root extract,LSE)对小鼠急性肝损伤的防护作用及药理机制。方法建立小鼠四氯化碳(CCl_4)肝损伤模型,检测LSE对血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)活性的影响;观察肝细胞超微结构的变化;测定肝线粒体膜电位以及线粒体对高钙诱发肿胀的敏感性的变化。结果一定剂量(100~400 mg/kg)LSE能有效抑制CCl_4引起的小鼠血清AST及ALT活性的显著上升;明显减轻肝损伤引起的小鼠肝脏线粒体形态结构的破坏:抑制线粒体膜电位的降低以及线粒体对外加高钙诱发肿胀敏感性的下降。结论LSE对CCl_4诱导的急性肝损伤具显著的防护作用,其机理可能与保护肝脏线粒体有关。 Objective To study the hepatoprotective effect of Limonium sinense (Girard) Ktze root extract (LSE) and the possible mechanisms underlying its protection on acute liver injury induced by carbon tetrachloride ( CC14 ). Methods CC14 - induced liver injury model was used to evaluate the effect of LSE on the activities of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in mice.' Uhrastructure of liver was observed and liver mitochondrial membrane potential and mitochondrial swelling were measured following CC14 injection without or with LSE. Results With oral pretreatment of 100 mg/kg, 200 mg/kg or 400 mg/kg LSE, the increase in serum AST and ALT activities induced by CC14 were obviously reversed and significant morphological changes in liver mitochondria were remarkably lessened. In addition, the decrease in mitochondrial membrane potential and sensitivity of mitochondrial swelling to the exotic Ca^2+ stimulation induced by CC14 were also prevented by LSE. Conclusion LSE has hepatoprotective activity and /he mechanisms underlying its protective effect may be related to its protection on liver mitochondria.
