摘要
通过金属离子及表面活性剂的单因素试验及正交试验,对Bacillus subtilisNX-21胞外合成γ-谷氨酰转肽酶的发酵条件进行优化。结果表明,Mn2+、Mo6+和Tween-20的使用可有效促进菌体产酶,其最佳组合配方为:Tween-20 0.15%、(NH4)2MoO40.8 g.L-1、MnSO40.05 g.L-1。优化后该菌株产酶酶活达到3.9×103U.L-1,较原培养条件提高了21.6%。
For the production of γ- glutamyhranspeptidase by Bacillus subtilis NX - 21, the conditions of metal ions and surfactant were investigated based on single factor and orthogonal experiments. It was shown that Mn^2+ , Mo^6+ and Tween -20 had remarkable positive effects on the production of γ-glutamyhranspeptidase. The opti- mized conditions were: Tween -20 0.15% , (NH4)2MoO4 0.8g · L^-1, MnSO4 0.05g · L^-1. γ - glutamyltranspeptidase activity was 3.9U · mL^-1 , which was 21.6% higher than the result of previous experiment.
