动脉粥样硬化病变中p53抑癌基因结构突变的研究

摘要 本文应用高灵敏、高特异的PCR-SSCP方法,首次系统地研究了中国成人AS病变组织中p53anti-oncogene的突变。结果发现:受检的89例AS组织中的9例有p53基因的突变(有PCR-SSCP的异常),其中6例经测序确定了p53基因突变的具体位点。结论:在人类的AS病变中确有p53anti-oncogene的突变,此可能为AS形成过程中新的主要机制之一。 Controlling by Chinese healthy adultsnormal artery cells and healthy adults normal blood lymphocytes(DNA), the mutations of p53 anti oncogene in 89 cases of Chinese adults atherosclerotic(AS) plaques were studied detailedly and systematically. The results showed that totally 9 cases(9/89) mutations of p53 gene were found by radiative PCR SSCP, a highest sensitive and highest specific method for the detection of DNA mutations, in which, 6 cases had been further verified by sequencing. In conclusion, from our studies, its clear that there are definite mutations of p53 anti oncogene in some Chinese adults AS plaque cells, and this phenomenon may be a new mechanism in the pathogenesis of human AS damages.
