摘要
本文应用高灵敏、高特异的PCR-SSCP方法,首次系统地研究了中国成人AS病变组织中p53anti-oncogene的突变。结果发现:受检的89例AS组织中的9例有p53基因的突变(有PCR-SSCP的异常),其中6例经测序确定了p53基因突变的具体位点。结论:在人类的AS病变中确有p53anti-oncogene的突变,此可能为AS形成过程中新的主要机制之一。
Controlling by Chinese healthy adultsnormal artery cells and healthy adults normal blood lymphocytes(DNA), the mutations of p53 anti oncogene in 89 cases of Chinese adults atherosclerotic(AS) plaques were studied detailedly and systematically. The results showed that totally 9 cases(9/89) mutations of p53 gene were found by radiative PCR SSCP, a highest sensitive and highest specific method for the detection of DNA mutations, in which, 6 cases had been further verified by sequencing. In conclusion, from our studies, its clear that there are definite mutations of p53 anti oncogene in some Chinese adults AS plaque cells, and this phenomenon may be a new mechanism in the pathogenesis of human AS damages.
