摘要
目的:观察热疗联合足叶乙甙增强对慢性髓系白血病细胞株K562的体外增殖的抑制作用及对凋亡的影响。方法:采用MTT法确定足叶乙甙的工作浓度,以该浓度进行化疗或与热疗的联合,选择温度40℃及42℃,体外作用于K562。作用前及48小时,采用台盼蓝拒染法检测肿瘤细胞的存活率,MTT法检测对肿瘤细胞增殖的抑制作用,流式细胞仪检测肿瘤细胞的凋亡。观察热疗联合足叶乙甙的抗肿瘤效果。结果:作用48小时IC50的药物浓度作为实验的工作浓度。单纯40℃、42℃热疗60分钟对K562细胞系有抑制作用(P<0.01),并随温度增高而增强;单纯化疗对K562细胞也有抑制作用;各热化疗组对K562有显著的抑制作用(P<0.01),随着温度的增高而增强。流式细胞仪检测细胞凋亡,热疗组、化疗组及热化疗组细胞凋亡率均较对照组显著升高,各组之间均有显著性差异(P<0.01)。结论:热疗联合足叶乙甙能增强对K562细胞的体外抑制作用,并可以提高肿瘤细胞的凋亡率。
Objective:To investigate the suppression effect and apoptosis of chronic leukemic cell line K562 treated with etoposide and hyperthermia. Methods : The working concentration of etoposide against K562 was determined by MTT assay. Then hyperthermia and chemotherapy were used singly or concurrently and the cell survival rates were obtained at 48h. The suppression effect was evaluated by MTT assay. The apoptosis rates of K562 were determined by flow cytometric analyses. Results:The concentration of etoposide in the experiment was defined as its IC50. The 40℃ and 42℃ hyperthennia for 60 min showed obvious inhibition to K562 at 48h ( P 〈0. 01 ) ,etoposide chemotherapy or hyperthermia both obviously inhibited the growth of K562 at 48h (P 〈0.01 ). Etoposide chemotherapy combined with hyperthermia showed obvious effect to K562. According to flow cytometric analyses, the treatment of hyperthermia and etoposide alone or concurrently could obviously increase the apoptosis rates of K562 ( P 〈 0. 01 ). ConcIusion: Etoposide chemotherapy combined with hyperthermia 60min showed obvious suppression effect to K562 at 48h. The synergism might increase apoptosis rates of K562.
