抑癌基因RASSF1在胃癌中的表达及临床意义 被引量:1

摘要 目的:探讨RASSF1抑癌基因二种不同转录本在胃癌中的表达情况及与临床的关系。方法:采用RT-PCR方法检测32例胃癌组织及相应的癌旁正常组织,以及51例胃良性病变中RASSF1A、RASSF1B mRNA的表达情况。结果:RASSF1A、RASSF1B在癌旁正常组织中表达缺失率为0,在胃良性病变中有表达缺失,在胃癌组织中的表达缺失率分别为59.4%(19/32)和28.1%(9/32)。RASSF1A、RASSF1B表达与胃癌分化程度及组织学类型无显著相关性。结论:RASSF1二种不同转录本RASSF1A、RASSF1B在癌旁正常组织中可完全表达,在胃癌中的表达缺失率不同,与胃癌的分化程度无明显相关。RASSF1A在胃癌中表达缺失明显,表明RASSF1A是一种具有抑癌作用的基因。 Objective:To investigation the expression lose rate of two kind different RASSF1 transcripts RASSF1 A, RASSF1B in gastric carcinoma and benign gastric disease and discuss their relations with clinical significance. Methods :The mRNA expression of RASSF1A and RASSF1B were detected by RT-PCR in 32 patients with gastric carcinoma, 32 adjacent non-carcinomatous gastric tissues and 51 case with benign gastric disease. Results.The rate of expression loss of transcripts A and B in adgastric carcinoma were 59. 4% and 28.1% , respectively. The rate of expression loss of transcripts A and B in adjacent non-carcinomatous were zero. RASSF1A and RASSF1 B mRNA expression have no statistical relation with different grade of gastric carcinoma. Conclusion:The two different RASSF 1 transcripts RASSFl A, RASSF1B are all expression in adjacent non-carcinomatous. RASSFl A is a significant expression loss in gastric carcinoma, and has no statistical relation with different grade of gastric carcinoma. The result indicates that RASSF1A is a suppressor gene.
