摘要
目的探讨TNF-α对去子宫大鼠卵巢颗粒细胞凋亡的影响。方法应用子宫切除术建立去子宫Sprague-Dawley大鼠模型。体外培养卵巢颗粒细胞并加60 ng/ml外源性TNF-α。应用荧光显微镜、流式细胞仪检测细胞凋亡情况。结果模型组较正常对照组凋亡增加;TNF-α组较模型组凋亡进一步加剧。结论外源性TNF-α促进去子宫大鼠卵巢颗粒细胞凋亡。提示TNF-α可能加速了子宫切除所致卵巢早衰过程。
Objective To investigate the influence of TNF -α on the apoptosis of ovary granulosa cells of rats with hysterectomy. Methods The rats model of Sprague - Dawley with hysterectomy were created by hysterectomy. Exogenesis TNF -α at concentration of 60ng/ml were put into cultured ovary granulosa cells of rats accepted hysterectomy. The apoptosis was detected by fluorescence microscope and flow cytometer. Results The apoptosis of cells in model group showed more than that in control group; The apoptosis of cells in TNF -α group were more than that in the model group. Conclusion Exogenesis TNF -α may accelerate the apoptosis of ovary granulosa cells and premature ovarian failure of rats with hysterectomy.
