摘要
目的:研究穿山龙总皂苷对佐剂性关节炎大鼠(AA)的免疫调节作用。方法:将大鼠随机分为对照组、阳性药物组和实验组。连续灌胃给药8天,停药后处死大鼠,计算胸腺指数、脾指数,观察血清和关节液IL-1、IL-6、IL-8、TNF-α水平,观察ConA诱导的脾细胞增殖反应。结果:不同剂量穿山龙总皂苷治疗能使AA大鼠降低的胸腺指数基本恢复到正常,也能使AA大鼠明显增大的脾指数显著降低。高、中、低剂量组给药大鼠的血清、关节液内炎性细胞因子IL-1、IL-6、IL-8、TNF-α水平较AA模型组明显降低(P<0.01),ConA诱导的脾细胞增殖反应明显提高。结论:穿山龙总皂苷有良好的抗炎及免疫调节作用,其机理部分可能与调节机能依赖性的双向免疫及抑制炎性细胞因子产生有关。
Objective :The immunoregulatory effect of whole saponin of Rhizoma Dioseoreae Nipponieae(RDN) on rat with adjuvant arthritis (AA) induced by Freund's complete adjuvant (FCA) were investigated. Methods : The 52 rats with AA were divided into the AA model group, the positive drug group and the experimental groups, the normal rats were divided into control group . The affected rats were given gastric perfusion of RDN. Results : At the 22th day of injecting FCA, the thymas index of AA model rats was decreased, and so did the normal rats treated with RDN. The AA rats treated with RDN increased their decreased thymas index. It showed that the RDN could have two - sided regulatory effects to AA. The levels of IL - 1, IL - 6, IL - 8 and TNF-α of the affected rats treated by RDN were remarkably diminished in serum and joint fluid than that of in AA model ( P 〈 0. 01 ). Conclusions : The results showed that RDN had a dramatically effect on anti -inflammation and immunoregulation. Its good therapeutic role in rats with AA may be associated with its two - sided immunoregutalion and inhibition to the production of IL - 1 ,IL -6,IL -8 and TNF-α.
