IgA肾病大鼠外周血、肾组织γδT细胞分泌TGF—β1功能的实验研究

摘要 目的研究IgA肾病大鼠外周血、肾组织γδT淋巴细胞分泌细胞因子TGF—β1的功能。方法流式细胞仪分析IgA肾病大鼠外周血、肾组织78T淋巴细胞比例以及血清IgA水平,并体外选择性扩增γδT细胞,在体外培养条件下,测定培养体系中细胞因子TGF—β1的表达。结果IGA肾病大鼠外周血、肾组织中γδT淋巴细胞比例升高,并能异常分泌TGF—β1细胞因子。结论γδT细胞可能是通过分泌TGF—β1参与IgA肾病的发生与进展。 objective To study TGF-β1 productions by stimulated γδT cells in peripheral blood mononuclear cells (PBMC) and the kidney in IgA nephropathy (IgAN) rat. Methods The proportion of γδT cells and the levels were of serum IgA measured by flow cytometry and ELISA, respectively. The solid phase monoclonal anti-TCRγδMc was used to isolated γδT cells . γδT cells were stimulated with anti-CD3 for 48h. The levels of TGF-β1 in culture supernatants of γδT cells were measured by ELISA. Results The proportions of γδT cells in PBMC and the kidney from IgAN rats were significantly higher than that in the controls. High purifiedγδT cells were obtained by stimulation of solid phase antibody. The levels of TGF-β1 produced by activated γδT cells from IgAN rat were significantly higher than those from the controls. Conclusion γδT cells might be play important role in pathogenesis of IgAN via the production of TGF-β1 .
