摘要
用SP2/0骨髓瘤细胞与经人促甲状腺激素(hTSH)免疫的BALB/C小鼠的脾细胞融合,融合率60.4%,抗体阳性率4.3%,经3~4次克隆化,筛出3株稳定分泌TSHMcAb的杂交瘤细胞株。用放射免疫分析法(RIA)检测小鼠腹水,抗体效价为10-4时,抗体结合率为45.5%。用ELISA方法检测小鼠腹水,抗体效价达10-5。其中3株细胞做琼脂糖免疫双扩散试验,均显示为IgG1,亚类。染色体计数为95对,证明为杂交瘤细胞。
SP2/0 myeloma cells are fused with spleen cells of BALB/C mice immunized with hTSH. The rate of fusion is 60.4%,and the positive rate of antibody is 4.3%. After cloning, 3 hybridoma cell lines of secretion hTSH McAb is established. When the activity of McAb of ascites diluted to 1∶10 -4 is tested by RIA, the binding rate is 45.5%.Ig class and subclass are determined with agar double diffusion test, 3 hybridoma cell lines secreting McAb are IgG 1 subclass.
