卡介苗联合药物缓释剂在抗肿瘤方面的初步研究 被引量:1

摘要 目的初步探讨卡介苗(BCG)联合药物缓释剂的抗肿瘤性。方法建立大鼠肿瘤动物模型并给予不同BCG处理,取新鲜胸腺组织,行胸腺细胞增殖和凋亡的流式细胞学检测,并取皮下肿瘤常规制备石蜡切片行HE染色,光镜观察肿瘤内的形态变化。结果①造模后7天、14天,注射BCG及BCG结合泊洛沙姆407(P407)组肿瘤组织坏死明显,肿瘤周围有明显的纤维化。②BCG与P407混合液处理组胸腺细胞凋亡率明显低于空白对照组,而增殖指数明显高于该组;BCG原液在造模后7天时,胸腺细胞凋亡率与空白对照组相比无显著差异,而增殖指数明显高于该组,造模后14天时,胸腺细胞凋亡率及增殖指数均明显高于正常对照组。结论P407可增强BCG的抗肿瘤作用。  Objective To explore the effects of slow-releasing reagents combined with Bacillus Calmette-Guerin vaccine(BCG) on anti-tumor.Methods After making the models with liver cancer,different reagents were injected into the bodies of the rats in corresponding group.The injected reagents in groups were BCG,BCG combined with gelatin,BCG and BCG combined with P407 and PBS.The gross changes in the region where reagent was injected were observed.Tissue was taken for further observation and studied by routine HE staining.Simultaneously,the thymus of rats was prepared for flow cytometric DNA analysis(FCM).Results(1)7 and 14 days after establishing models,the obvious necrosis could be found in the tumor of BCG and BCG combined with P407 treated group,and there was significant fibrogenesis around the tumor.(2)FCM analysis revealed that 7 and 14 days after establishing models,the apoptosis rate of thymocyte in BCG combined with P407 group was much lower than that in the controls,and its proliferation index was much higher.After 7 days,the proliferation index of thymocyte in BCG group was much higher than that in the controls while there was no significant difference in the apoptosis rate of thymocyte between these two groups after establishing models for 14 days, the apoptosis rate and the proliferation index in BCG treated group were both higher than that in the controls.Conclusion P407 can enhance the effect of BCG on anti-tumor.
