补肾固精方治疗BXSB小鼠狼疮性肾炎的实验研究 被引量:3

摘要 目的:探讨中药补肾固精方对BXSB小鼠狼疮性肾炎的治疗作用。方法:BXSB小鼠随机分为模型组(生理盐水)、中药组(补肾固精方)、激素组(地塞米松)与中西药结合组(补肾固精方与地塞米松合用)。各组均连续用药45d,于第46天取材。考马斯亮蓝G250法检测尿蛋白定量,ELISA法测定血清抗ds-DNA抗体,HE染色光镜下观察肾脏病理改变,直接免疫荧光法观察肾组织中IgG沉积。结果:与模型组比较,中药组尿蛋白定量显著减少,血清抗ds-DNA抗体显著降低,免疫复合物沉积减少,肾脏病理损害有所改善;中西药结合组与模型组相比有显著差异,且优于中药组与激素组。结论:补肾固精方可以降低尿蛋白定量和血清抗ds-DNA抗体水平,改善肾脏损害,其与激素有协同作用。 Objective: To Study on the effect of "Bushen Gujing Decoction" on lupus nephritis. Methods: BXSB mice were randomly divided into four groups: model group, "Bushen Gujing" group, dexamethasone group, and "Bushen Gujing" plus dexamethasone (BGD) group, and were continuously treated for 45 days. Proteinuria was detected by Coomarssie brilliant blue G250 method, and the level of serum anti ds-DNA antibody was assayed by ELISA. The sediment of lgG immune complex in kidney was determined by direct immunofluorescence, and pathological damage of kidney was determined by HE staining method. Resuits: Compared with model group, the levels of anti-ds-DNA antibody, 24 h urine protein and lgG sediment in "Bushen Gujing" group decreased significantly, and the histomorphological injury of kidney in "Bushen Gujing" group was also improved. The efficacy in BGD group was better than in "Bushen Gujing" group and dexmethasone group. Conclusion: "Bushen Gujing Decoction" may decrease proteinuria, and the level of anti-ds-DNA antibody, improve the damage of kidney of BXSB mice. "Bushen Gujing Decoction" combined with steroid can enhance the effect.
