摘要
目的观察骨髓间充质干细胞移植对大鼠大脑中动脉栓塞后神经功能的影响及GDNF的表达。方法采用线栓法大鼠大脑中动脉栓塞(MCAO)模型,颈内动脉移植MSCs,Bederson神经功能评分观察大鼠神经功能,免疫组织化学和原位杂交技术检测GDNF表达。结果MSCs移植可显著提高大鼠局灶性脑缺血后神经功能,缺血周边区GDNF蛋白及mRNA表达水平均高于对照组。结论MSCs移植可促进神经功能恢复,可能通过增加GDNF的表达起到脑保护作用。
Objective To observe tile neurological functional recovery after transplanting mesencbymal stem cells (MSCs) to rats with middle cerebral artery occlusion (MCAO), and detect the expression of GDNF. Methods MCAO model was established with suture emboli method, measure functional outcome with Bederson neurological severity score. MSCs were transfered into rats through arteria carotis internal 24 hours after MCAO, detect expression of GDNF protein and mRNA by immunohistochemieal and in situ hybridiza- tion method. Results MSCs transplantation could significantly improve the rats neurological functional recovery after MCAO. The expression of GDNFmRNA and protein is higher than the control group. Conclusion MSCs can significantly improve neurological recovery after MCAO in rats and protect the brain by increased GDNF expression.
