乙酰水杨酸对大鼠局灶性脑梗死周边神经细胞细胞周期素表达的影响

摘要 目的:观察成年大鼠局灶性脑梗死后神经元内细胞周期素D1和细胞周期素B1的表达以及乙酰水杨酸对其的影响。方法:线栓法制作大鼠大脑中动脉永久闭塞模型。40只大鼠随机分为假手术组(n=8)、缺血组(n=16)、乙酰水杨酸干预缺血组(n=16)。采用HE染色显示梗死灶,应用免疫组化方法观察各组大鼠脑内细胞周期素D1和细胞周期素B1阳性染色神经细胞的数量和分布。结果:HE染色显示,缺血组大鼠缺血灶侧皮质可见不规则坏死区,乙酰水杨酸干预组坏死灶较缺血组显著缩小。缺血灶周围区有较多细胞周期素D1和细胞周期素B1阳性神经元分布,但细胞周期素B1阳性细胞数显著少于细胞周期素D1阳性细胞数,乙酰水杨酸组显著少于缺血组(P〈0.01)。乙酰水杨酸组细胞周期素D1阳性神经元多呈胞质阳性,而缺血组多为胞核阳性。结论:成年大鼠局灶性梗死周边神经元内异常表达细胞周期素D1和细胞周期素B1,乙酰水杨酸可以减少细胞周期素D1和细胞周期素B1表达,进而缩小梗死病灶。乙酰水杨酸还可能抑制细胞周期素D1由胞质向胞核转运。 Objective: To observe the expressions of cyclin D1 and cyclin B1 in the tissues around the cerebral infarction in rats and the effect of acetyl salicylic acid on them. Methods: A model of permanent focal ischemia was induced by the intraluminal filament occlusion of the middle cerebral artery (MCAO) in rats. Forty rats were randomly allocated into sham-operation (n =8), ischemia (n = 16) and acetyl salicylic acid intervention (n = 16) groups, qqae infarction foci were revealed by H&E. staining, The numbers of positive-staining neurons and the distribution of cyclin D1 and cyclin B1 were observed in all groups by using immunohistochemistry. Results: H&E staining showed that the irregular necrotic areas in the cortexes on the ischemic sides were observed in the ischemia group, The necrotic foci in the acetyl salicylic acid group reduced significantly as compared with the ischemia group. There were more cyclin D1 and cyclin B1 positive neurons distribution around the ischemic foci, but the numbers of cyclin B1 positive neurons were significantly less than those of the cyclin D1; the acetyl salicylic acid intervention group was significanly less than the ischemia group (P 〈0.01). Most of the cyclin D1 positive neurons were cytoplasm-positive in the aspirin intervention group, but most of them were nuclear positive in the ischemia group. Conclusions: qhe cyclinD1 and cyclin B1 in the tissues around the cerebral infarction were expressed abnormally in rats. Acetyl salicylic acid may reduce the expressions of cyclin D1 and cyclin B1, and thus it reduces infarction volume Acetyl salicylic acid may also inhibit the transportation of cyclin D1 from cytoplasm to nuclear.
