摘要
分析了影响牛体外成熟卵母细胞电激活的主要参数,初步建立了电激活的实验程序。结果表明,卵母细胞激活率随着卵龄的延长、电场强度的增加以及电脉冲次数的增多而升高,含Ca2+、Mg2+离子的电激液显著高于非电解质液。以0.3mol/L甘露醇+100μmol/LCaCl2+100μmol/LMgCl2为电激液,施加1.6kV/cm、160μs,3次电脉冲间隔15min,激活率、卵裂率、囊胚发育率分别为86.0%,86.5%及48.9%
In this paper, the factors affecting electrical pulsing activation were analyzed, and the experimental procedure for activation was established. The results indicated that the electro activation rate became higher with the increase of the oocyte age,field strength and pulse times, and also significantly higher in the electrolyte medium than that in nonelectrolyte medium.When the oocytes were pulsed in the medium of 0.3 mol/L mannitol+100 μmol/L CaCl 2 +100 μmol/L MgCl 2 with the pulsing parameters:duration of 160 μs, field strength of 1.6 kV/cm, 3 pulses at 15 min intervals, the activation rate, cleavage rate,developmental rate of blastocyst were 86.0%, 86.5% and 48.9% respectively.
