摘要
目的探讨亚低温对Nogo-A和生长相关蛋白(GAP-43)表达的影响,为亚低温的临床应用提供理论依据。方法将实验动物分为正常对照组、亚低温干预组、室温恢复组,应用免疫组化方法检测标本中Nogo-A和GAP-43蛋白,比较各组标本阳性细胞的个数,初步探讨亚低温对Nogo-A和GAP-43表达的影响。结果亚低温干预组与室温恢复组相比,qNogo-A(Ⅲ,Ⅱ)=8.83,qgap-43(Ⅱ,Ⅲ)=11.65,差异均有统计学意义(P<0.01)。而室温恢复组又与正常对照组相比,qNogo-A(Ⅲ,Ⅰ)=11.16,qgap-43(Ⅰ,Ⅲ)=13.98,差异也均有统计学意义(P<0.01)。结论亚低温干预可抑制Nogo-A的表达,维持接近正常的GAP-43表达,有利于新生动物缺氧缺血性脑损伤早期神经元的再生。
Objective To study the effect of sub-hypothermia on the expressions of Nogo-A and growth-associated protein-43 (GAP-43), and to provide theoretic references for its clinical application. Methods Neonatal rabbits were divided into control group, sub-hypothermia group and room tempera- ture group randomly. Protein Nogo-A and GAP-43 of each specimen were measured by immunohistoehem- ical assays and the number of positive cell was calculated and compared among groups in order to study preliminarily the effects on gene expression of Nogo-A and GAP-43 after sub-hypothermia treatment. Re- suits Comparing the average number of positive cells of Nogo-A and GAP-43, sub-hypothermia group and control group had significant difference with room temperature group (qnogo-A (m, n) = 8.83, qgap-43( Ⅱ/, Ⅲ)=11.65, P〈0.01; qNogo-A(Ⅱ,I) = 11. 16, qgap-43(Ⅰ,Ⅲ) =13.98, P〈0.01). Conclusions Sub-hypo- thermia can inhibit Nogo-A expression and maintain the expression of GAP-43 at a near normal level, which will be benefit the regeneration of neurons in neonatal rabbits suffering from hypoxie-isehemie brain damage.
