摘要
目的 分析亚硝酸盐定量检测的重要方法-α-萘胺显色反应的可靠性.方法 弱酸条件下,亚硝酸盐与对氨基苯磺酸重氮化后,再与α-萘胺偶合显色后分光光度法测定亚硝酸盐氮含量.结果 该法检出限为0.5 ng/ml,亚硝酸盐氮浓度在(4~400)ng/ml范围内有良好的线性关系,相关系数 r=0.999 9(n=10 ).回收率96.8%,RSD为1.86%.亚硝酸盐氮浓度大于40 μg/ml时无显色现象.结论 该方法灵敏度高,选择性好,简便实用,在一定浓度范围内,结果准确可靠,应用时需注意控制,防止假阴性错误,以及假阳性干扰.
Objective To analyze the reliability of α - naphthylamine color reaction for determining nitrite. Methods Nitrite was diazotized by sulfanilic acid in the condition of feeble acid, and then coupled with α - naphthylamine to form a purple dye which was determined by spectrophotometry. Results The limit of detection is 0.5ng/ml, and the linear range of nitrogen in nitrite is from 4ng/ml to 400ng/ml ( r = 0.9999,n = 10 ). The recovery is 96.8% and RSD is 1.86%. It showed that there was no purple dye to form when the concentration of nitrogen in nitrite was above 40μg/ml. Conclusion This method is high sensitive and selective, and it is practical. Its results are exact in a range of concentration. It is important to control the condition of reaction well in order to avoid falsity of negative or positive.
