摘要
目的研究E-钙黏附素(E-cadherin,E-cad)和多药耐药P-糖蛋白(P-gp)、谷胱甘肽转移酶(GST-π)、拓扑异构酶(TopoⅡ)在大肠癌中的表达,探讨E-cad与大肠癌耐药的关系。方法应用免疫组化S-P法联合检测E-cadherin、P-gp、GST-π、TopoⅡ在84例大肠癌组织和20例正常大肠黏膜组织中的表达,并结合临床病理因素进行分析。结果正常大肠黏膜上皮细胞中E-cad均呈阳性表达,大肠癌组织中E-cad的阳性表达率为43.3%;高、中分化腺癌E-cad阳性表达率显著高于低分化腺癌(P<0.05);E-cad表达阳性组P-gp、GST-π显著高于E-cad表达阴性组(P<0.05),而E-cad表达阳性组TopoⅡ表达则显著低于E-cad表达阴性组(P<0.05)。结论表明E-cad表达与大肠癌原发性耐药相关。
Objective To study the expression of E-cadherin, P-glycoprotein(P-gp), glutathione-s-trannsferase (GST-π) and DNA topoisomerase(Topo Ⅱ ) in adenocareinoma of the colorectal tissue, and to explore the relationship of E-cadherin and drug-resistance in colorectal tumor. Methods The expression of E-cadherin, P-gp, GST-π and TopoⅡ in 84 cases of colorectal adenocarcinoma and 20 cases of normal tissues were detected by S-P immunohistochemical technique, and its correlation to clinical pathologic characteristics of carcinoma was explored. Results The expression rate of E-cadherin in colorectal carcinoma was lower than that in normal tissues(P〈0. 05). The expression rates of E-cadherin in group of highly and moderately differentiated adencarcinoma were significantly higher than those in group of poorly differentiated adenocarcinoma(P〈0.05). The expression of P-gp and GST-πin group of E-cadherin+ were higher than those in group of E-cadherin(P〉0.05) ;but the expression of Topo Ⅱ in group of E-cadherin+ were lower than those in group of E-cadherin- (P〈0.05). Conclusion Expression rates of E-cadherin is association with the primary drug-resistance in colorectal carcinoma.
