RP-HPLC法测定蒲地蓝消炎片中咖啡酸的含量研究

Determination of Coffee Acid in Pudilanxiaoyan Tablet with RP-HPLC Method

目的：建立用反相高效液相色谱（RP-HPLC）法测定蒲地蓝消炎片中咖啡酸含量测定的方法。方法：采用RP-HPLC法进行测定,色谱柱为Dikma C18（250 mm×4.6 mm,5μm）;流动相：甲醇-磷酸盐缓冲液（取磷酸二氢钠1.56 g,加水使溶解成1000 ml,再加1%磷酸溶液调节pH值至3.8~4.0）;检测波长为323 nm;柱温：40℃。结果：咖啡酸在一定色谱条件下能够完全分离,咖啡酸线性范围8.19~49.14μg.ml^-1,平均加样回收率分别为99.89%;RSD%为1.27%。结论：本方法可有效地测定蒲地蓝消炎片中咖啡酸的含量。

Objective： To establish RP - HPLC method for determination of coffee acid in Pudilanxiaoyan Tablet. Method： The chromatography consisted of chromatograph volume of DikmaC18 （250 mm × 4.6 mm, 5 μm） with flow phase of methanol - buffer phosphate（taking natrium biphosphoricum 1.56g to dissolve into water about 1000 ml, and again adding 1% phosphoric acid to adjust pH value in 3.8 ~4.0）; the detection wavelength was 323 nm, column temperature was 40 ℃. Results： Coffee acid could be isolated completely at a certain chromatographic condition, the linear range of coffee acid was 8.19 ~ 49.14μgoml - 1, The average recovery was 99.89% with RSD% of 1.27%. Conclusion： The method can determine the content of coffee acid in Pudilanxiaoyan Tablet accurately.
