摘要
目的:建立人血浆中罗红霉素的高压液相/质谱(HPLC-MS)测定方法。方法:采用Diamon- sil-C 18柱(200mm×4.6mm,5μm),流动相为甲醇(10mmol/L)醋酸铵缓冲液(pH=3.5)80:20,流速为1.0ml/min。用含内标(克拉霉素)的乙腈液沉淀蛋白,离心后取上清液进样,以质谱为检测手段,按内标法定量。结果:本法在0.05~10.0μg/mL范围内线性关系良好,批内RSD<1.96%,批间RSD<2.50%,批内RE<4.96%,批间RE<5.54%。结论:该方法灵敏度高,精密度和准确度好,适用于罗红霉素血药浓度的测定。
Objective: To establish a HPLC-MSmethod for determining the concentration ofroxithromycin in plasma. Methods: This assay is conducted on Diamonsil C 18 column(200 mm× 4.6 mm, 5 μm). The mobile phase consists of methanol(10 mmol/L)and ammonium acetate(pH=3.5)80:20, at the flow-rate of 1.0 ml/min. The plasma was treated with acetonitrile, which contains clarithromycin. After the centrifuge of this mixture, the supernatant was injected into the chromatographic system. This method adopts MS detectorand is based onthe internal standardto analyse quantitatively. Results: Roxithromycin has good linear relationship between 0.05- 10 μg/mL. Within-group and between-group precision expressed by relative standard deviation is less than 1.96% and 2.50%. Intra-group and inter-group accuracy expressed by RE is less than 4.96% and 5.54%. Conclusion: The method is highly sensitive, precise and accurate. It is applicable to the determination of roxithromycin inplasma.
