喉癌MMP-2和TIMP-2表达与瘤内微血管密度的相关性 被引量:4

摘要 目的对喉癌组织中MMP-2、TIMP-2的表达与瘤内微血管密度(microvessel density,MVD)的相关性及预后意义进行探讨。方法对37例喉鳞癌患者的组织切片进行MMP-2、TIMP-2及CD34的免疫组化检测,用计算机图像分析系统对组织切片结果进行测定。结果淋巴结转移组MMP-2表达水平比非淋巴结转移组明显升高(P<0.01),而TIMP-2的表达两组之间没有统计学差异(P>0.05)。MVD均值转移组比非转移组明显高(P<0.01),MVD值与MMP-2表达水平之间呈线性相关(P<0.05)。并发现高MVD值组MMP-2的表达明显高于低MVD值组(P<0.05),高MMP-2表达组无瘤生存期分别比低MMP-2表达组明显缩短(P<0.01)。结论MMP-2可能在喉癌侵袭、转移和血管生成中承担着一个重要角色,并且MMP-2的表达可能促进了喉癌的淋巴播散能力,有望成为喉癌患者的一个预后目标。  OBJECTIVE To understand the function of MMP-2 and TIMP-2 and the relationship among expression of MMP-2, TIMP-2 and microvessel density in laryngeal carcinomas. METHODS The expression of MMP-2, TIMP-2 and CD34 in 37 laryngeal carcinomas patients were studied with immunohistochemical staining. The staining results were studied morphometrically with computer image analysis. RESULTS The mean values of MMP-2 expression and microvessel density (MVD) in squamous cell carcinoma group with lymph node metastasis were significantly higher than that without lymph node metastasis (P〈0.01). And the level of MMP-2 expression tended to increase accordingly with the clinical stage of the tumor. The linear correlation was found between the expression of MMP-2 and MVD (P〈0.05). All the patients had been followed-up for 26~42 months. The diseasefree survival interval in the higher MMP-2 expression group was significantly shorter than that in the lower MMP-2 expression groups. CONCLUSION The results suggest that MMP-2 expression may be of prognostic significance in laryngeal carcinomas, and the linear correlation between MVD and MMP-2 expression also indicate the important role of MMPs in angiogenesis.
