摘要
目的探讨Jun激活区域-连接蛋白1(c-Jun activation domain-binding protein1,Jab1)和p27kip1的表达与喉鳞状细胞癌(laryngeal squamous cell carcinomas,LSCC)发生、发展的关系及两种蛋白之间的相关性。方法免疫组织化学SP法检测23例正常喉黏膜(normal laryngeal tissues,NLT)和68例LSCC组织中Jab1和p27kip1的表达。结果NLT和LSCC组织中,Jab1和p27kip1蛋白阳性表达率差异均有显著性(P<0.01)。LSCC中Jab1的过表达与临床分期及颈淋巴结转移显著相关(P<0.05),p27kip1的表达与颈淋巴结转移显著相关(P<0.05),Jab1的表达与p27kip1的表达呈负相关(P<0.05)。生存率分析表明,Jab1的过表达与患者的总生存率呈负相关(P<0.05)。结论Jab1可能通过对周期素依赖性蛋白激酶抑制因子(cell cycle inhibitory protein,CKI)p27kip1蛋白的降解而参与LSCC的发生、发展,联合检测Jab1及p27kip1蛋白的表达有助于综合评估LSCC的生物学行为及预后。
OBJECTIVE To study the relationship of the expression of Jab1 and p27kip1 with carcinogenesis and development of laryngeal squamous cell carcinoma (LSCC) , and whether Jab1 expression is correlated with p27kip1 level. METHODS The expression of Jab1 and p27kip1 protein was examined by SP immunohistochemistry method in 23 specimens of normal laryngeal tissues and 68 specimens of LSCC. RESULTS The positive rates of Jab1 in NTL and LSCC were 13.04 % (3/23) and 52.94 % (36/68) respectively, with a significant difference (P〈0.01) between the two groups. The p27kip1 protein was expressed in 82.61% (19/23) of NTL and 41.18 % (28/68) of LSCC respectively, showing a significant difference (P〈0.01) . Jab1 overexpression was significantly associated with clinical stage and lymph node metastasis (P〈0.05). The p27kip1 protein expression was significantly associated with lymph node metastasis (P〈0.05) . Jab1 expression was negatively associated with p27kip1 expression (P〈0.05). Survival analysis revealed that Jab1 overexpression was negatively associated with overall survival significantly. Jab1 (+) /p27kip1 (-) revealed poorer overall survival (P〈0.05) . Jab1 (+) /p27kip1 (-) /lymph node (+) revealed the worst overall survival (P〈0.01) . CONCLUSION Jab1 protein may contribute to the carcinogenesis and development of LSCC by facilitating the degradation of cyclindependent kinase inhibitor (CKI) p27kip1. Combined detection of the expression of Jab1 and p27kip1 may be useful in evaluating the biological behavior of LSCC comprehensively.
