摘要
目的研究他莫昔芬(TAM)对人肝癌HepG2细胞增殖和雌激素受体(ER)表达的影响。方法将7.5、15、30μmol/L的TAM分别作用于HepG2细胞(观察组),对照组不加TAM,每孔加入1×105/ml细胞0.1 ml,培养24、48、72 h,采用MTT法测定HepG2抑制率;细胞免疫组化染色观察HepG2细胞ER的表达。结果TAM作用后HepG2细胞增殖随浓度增加及处理时间延长,细胞抑制率增加。结论TAM可抑制肝癌细胞增殖,其机理可能为降低ER表达。
[ Objective] To study effects of tamoxifen on proliferation and estrogen(ER) expression of human hepatoeellular carcinoma HepG2 cells. [ Methods] HepG2 cells were treated with tamoxifen, at concentration of 7.5 μmol/L, 15 μmol/L and 30 μmol/L, control group wasnt treated with tamoxifen. Then the cells were cultured for 24 h,48 h and 72 h respectively. MTY was used to determine suppression rate of HepG2 cells; cellular immunohistoehemistry was adopted to observe the effects of tamoxifen on ER expression of HepG2 cells. [ Results] Tamoxifen inhibited the proliferating rates of HepG2 cells in doesand time-dependent manner. Tamoxifen suppressed the ER expression of HepG2 cell in does- and time- dependent. [ Conclusions ] Tamoxffen can suppress human hepatocellular carcinoma HepG2 cells proliferation through inhibiting ER expression.
