GATA-3 and Differentiation of the Th1/Th2 Cells

摘要 机体清除病原体需要辅助性T细胞的参与,根据其产生细胞因子的不同可将辅助性T细胞分为两类:Th1细胞和Th2细胞。GATA3在调节Th1和Th2细胞分化中起重要作用。转录因子GATA家族(GATA-3)不仅在转录水平上调节Th2类细胞因子的表达,而且在染色体结构的重组和IL-4位点的开放方面都起着重要的作用。作为一个重要的调控,GATA3通过两种方式使Th2细胞表型稳定,首先,GATA3通过抑制IL-12受体β2链的表达来限制Th1细胞的分化;其次,GATA3通过正反馈调节来加强自身的表达。  T helper cells is needed in clearing pathogen of the body,which can be classified into Th1 and Th2 two distinct subsets according to different cytokines that they produced.GATA-3 plays a central role in regulating the differentiation of Th1 and Th2 cells.GATA-3 not only regulates Th2 cytokine expression at the transcriptional level,but also play an important role in recombination of chromosome structure and opening of IL-4 locus.As an important regulation factor,GATA-3 can stabilize the Th2 phenotype by two methods,of which the first one is to restrict the differentiation of Th1 by inhibiting the expression of β2-chain of IL-2 receptor,the second one is to strengthen its own exptession by positive feedback regulation.
