胸苷磷酸化酶与大肠癌肿瘤微血管密度及细胞凋亡的关系

摘要 目的探讨胸苷磷酸化酶(TP)/血小板衍生内皮细胞生长因子(PD-ECGF)与微血管密度(MVD)及大肠癌细胞凋亡的关系。方法应用免疫组化SP法,检测50例大肠癌组织中TP/PD-ECGF蛋白表达及微血管密度,采用TUNEL法检测大肠癌细胞凋亡指数AI,分析TP/PD-ECGF与MVD及大肠癌细胞凋亡的关系。结果TP/PD-ECGF表达强度和MVD两者呈正相关(r=0.72),与细胞凋亡指数呈负相关(r=-0.82),TP/PD-ECGF阳性组预后差。结论TP/PD-ECGF与大肠癌血管生成密切相关,对大肠癌的生长与浸润转移起促进作用,同时可抑制肿瘤细胞凋亡。TP/PD-ECGF与MVD可作为反映大肠癌生物学行为的指标,同时也是判断预后和指导辅助治疗的有效指标。 Objective To study the relationship between the thymidine phosphorylase(TP)/platelet-derived endothelial cell growth factor(PD-ECGF) and the microvessel density (MVD) and apoptosis index (AI) of colorectal carcinoma. Methods TP/PD-ECGF protein expression and MVD in 50 colorectal carcinomas were examined by means of immunohistochemical staining S-P method and AI of colorectal cancer cell was examined by TdT-mediated dUTP-biotin nick end labeling (TUNEL) method, the relationship between the TP/PD- ECGF,MVD and AI and prognosis of colorectal cancers were analysed. Results TP/PD-ECGF was negatively correlated with AI expression. There was a positive correlation between MVD and TP/P D-ECGF expression (r = 0.72 ). TP/PD-ECGF positive group had a bad prognosis. Conclusion TP/PD-ECGF is highly related to angiogenesis of colorectal carcinoma and promotes growth invasion and metastasis and repressed the cancer cell apoptosis . TP/PD-ECGF expression or MVD may be one of the predictors of the biological behaviors of colorectal carcinoma ,and they may serve as the prognostic factors and guide the decision on the therapy.
