卵巢肿瘤细胞DNA倍体分析及Fas表达研究

摘要 目的:通过检测卵巢肿瘤细胞DNA倍体及Fas蛋白表达情况,探讨其临床意义及其于卵巢细胞增殖与凋亡的关系。方法:应用流式细胞术(FCM)和荧光免疫技术,对34例卵巢癌组织和19例良性卵巢肿瘤及9例正常卵巢组织的卵巢细胞进行DNA倍体分析及Fas蛋白的表达测定。结果:①卵巢癌患者组中,卵巢组织细胞内DNA异倍体率明显高于良性卵巢肿瘤患者组及正常组。②卵巢癌异倍体率与组织学分级呈正相关。③卵巢癌Fas表达低于良性卵巢肿瘤。④卵巢癌Fas表达与临床分期组织学分级无关。结论:应用FCM进行卵巢癌细胞中的DNA倍体分析在临床上具有重要的诊断价值。卵巢癌组织中Fas表达下调与肿瘤细胞长期存活有关。 Objective: To detect the expression of DNA ploid and Fas in human ovarian tumor tissue, and explore the clinical significance and the relationship between the significance of proliferation and apoptosis of ovarian tumor cells. Methods: The expression of Fas and DNA ploid in 34 cases of ovarian tumor tissues, 19 ovarian benign tumor tissues and 9 normal ovarian tissues were detected with flow cytometry and immunofluorescence staining technique. Results: The DNA aneuploidy rate in ovarian cancer was higher than those in benign tumor and normal ovarian tissues. The DNA aneuploidy rate was correlated with histological grades. Fas was less expressed in ovarian cancer than that in benign tumor and nomal ovarian tissues. The expression of Fas was not correlated with histological grades. Conclusion: DNA aneuploidy expression has diagnostic value in ovarian cancer. The decreased Fas expressions in human ovarian cancer tissue are probably related to long survival of tumor cells.
