THE EXPRESSION AND SIGNIFICANCE OF Her-2 AND PTEN PROTEINS IN BLADDER CARCINOMA

摘要 目的研究Her-2和PTEN蛋白在膀胱癌中的表达及生物学意义。方法采用免疫组织化学SP法检测90例膀胱移行细胞癌(BTCC)和20例正常膀胱组织中Her-2、PTEN蛋白的表达。结果Her-2蛋白在正常膀胱组织中呈阴性表达,PTEN蛋白呈阳性表达;在90例BTCC组织中,Her-2、PTEN蛋白均呈不同程度表达;随肿瘤病理分级、临床分期增高,Her-2蛋白表达显著增强,PTEN蛋白降低(P值均小于0.01);在Her-2蛋白表达阳性、PTEN蛋白表达阴性病例中,随着肿瘤病理分级、临床分期增高,5年复发率均增高,各组间比较,差异均有显著性意义(P<0.05)。结论Her-2、PTEN蛋白表达检测可作为膀胱肿瘤的诊断指标之一,Her-2蛋白高表达是膀胱癌预后不良的指征,PTEN蛋白的异常表达可能成为一种新的预后判断指标。 Objective To investigate the expression and the biological significance of Her-2 and PTEN pro- teins in human bladder carcinoma. Methods Immunohistochemical S-P method was used to detect the expression of these two proteins in 90 cases of transitional cell carcinoma (BTCC) and 20 cases of normal bladder mucosa as a control group. Results The expression of Her - 2 was negative and the expression of PTEN was positive in control group, but Her -2 and PTEN appeared to be more different in BTCC, the positive expression of Her -2 increased with tumor grade increasing while PTEN decreased ( P 〈 0.01, respectively) ;the positive expression of Her - 2 increased with tumor stage increasing while PTEN decreased (P 〈 0.01, respectively) ;In cases which the expression of Her - 2 were positive and PTEN were negative, both recurrences of 5 years increased ( P 〈 0.05, respectively). Conclusion Her - 2 and PTEN might be regarded as an diagnosis marker of BTCC . The high expression of Her - 2 might be regarded as an indication of prognosis in bladder carcinoma. The PTEN may be a novel prognostic factor in BTCC .
