摘要
目的:建立以高效液相色谱法测定胸腺五肽(TP-5)含量的方法。方法:色谱柱为Kromasil C18,流动相为12%乙腈(含0.1%三氟乙酸),检测波长为222nm,流速为1.5mL.min-1,柱温为30℃,进样量为20μL。结果:TP-5检测浓度的线性范围为100~400μg.mL-1(r=0.9998);平均回收率为100.61%,RSD=0.46%。结论:本方法操作简便、灵敏、准确,可用于TP-5的含量检测。
OBJECTIVE: To develop a HPLC method for the determination of the content of thymopentin(TP - 5) . METHODS: Thymopentin was separated on Kromasil C18 with column temperature set at 30℃ .The mobile phase consisted of 12% acetonitrile (containing 0.1% trifluoroacetic acid) with flow rate of 1.5mL·min^-1 .The wavelength was set at 222nm and the sample size was 20μL. RESULTS: The standard curve of thymopentin was linear within the range 100-400μg·mL^-1 (r = 0.999 8),with average recovery at 100.61% and precision at 0.46%. CONCLUSION:This method is convenient, sensitive and accurate,and it can be used for determination of thymopentin.
