富硒黑木耳中硒多糖提取分离工艺的优化 被引量:8

摘要 以富硒黑木耳为原料,采用水浸提、乙醇沉淀多糖、Sevage脱蛋白、干燥等方法对富硒黑木耳中硒多糖的提取分离工艺进行了初步研究,并用蒽酮比色法和原子吸收法对多糖含量、硒含量分别进行了检测。结果表明,富硒黑木耳含硒多糖最佳提取条件为在料液比1∶20、提取时间2 h、提取温度60℃条件下提取两次,此时多糖提取率最高,水溶性多糖含量为18.95%,硒的含量为32.56μg.g-1。 In this article Auricularia auricula enriched selenium is used as experimental material to study effect of temperature,proportion and time on extraction of Se-polysaccharide by orthogonal experiment. The separation procedure of precipitation with alcohol and the purification from the removing proteins were deeply analyzed. Meanwhile the contents of polysaccharides were determined by Anthrone colorimetry. The results showed that the methods of the optimum parameters to extract soluble polysaccharides in A uricularia auricula were as follows: the ration between material and solvent is 1:20, the time is 2h, the temperature is 60℃. The content of soluble Se-polysaccharides is18.95%,the content of Se is 32.56μg·g^-1.
